Microglia are the rapid-response innate immune cells that nurture and protect the brain. These cells keep the brain in pristine condition for neuronal health by engulfing debris, toxins, and infections, and by supporting and rebuilding damaged synapses. As we age, these cells can become unfit to fulfil these functions, and a self-perpetuating cycle of reduced toxin clearance and neuroinflammation is sustained over the decades from mid-life to old age, often leading to Alzheimer's disease and related dementias. I am working to develop monoclonal and engineered antibody therapies as well as small molecule drugs to stimulate these immune cells. Our aim is to disrupt this neuroinflammatory cycle to prevent widespread neuronal loss and the onset of dementia. Specifically, my work involves:-recombinant receptor expression and purification-monoclonal antibody panning, production and characterisation-multibody engineering-tissue culture-structural biology-small molecule ligand screening-in vivo assay design in unique humanised AD models-ex vivo tissue examination-data analysis, interpretation and publication
