In my Post-doctoral fellowship, I am exploring about the biochemical studies of some of the mutants of HCM/DCM and its comparison with the wild type sarcomeric proteins (e.g. tropomyosin, troponin etc) by using various techniques like invitro motility assay, ATPase assay etc.

Title of PhD Thesis: Role of Voltage Dependent Anion Channel (VDAC)-Hexokinase Interaction in programmed cell death. In my PhD thesis, I have explored about,1. Biophysical characterization of VDAC purified from different sources by using Circular dichroism (CD), liposome swelling assay and and Electrophysiological characterization by bilayer lipid membrane (BLM) technique. For the purification of VDAC proteins I used various molecular biological techniques like DNA manipulation, sub-cloning, PCR, agarose electrophoresis, protein purification by HPLC based size exclusion, ion exchange and affinity chromatography, western blot.2. Explored the VDAC-Hexokinase interaction in animal and plants systems. I have used various cell biological approach like sub-culturing of mammalian cell culture (HeLa, HEK, CHO, and NFF cells), plant cell culture (BY2 cell line), and immunological techniques like Immuno-precipitation, Immunostaining, immunoblotting. I have used many microscopy like Fluorescence, Confocal and Total Internal Reflection Fluorescence (TIRF) microscopy to determine the VDAC-Hexokinase interaction in these system.
