The project was to: -Characterize a population of adrenocortical cancer cell line resistant to chemotherapy as a cancer stem cell-like cell line by identifying cancer stem cells properties such as high migration rate, increased invasiveness, protein expression of cancer stem cell markers -Understand the mechanism of endocytosis of the transporter highly expressed responsible for this chemotherapy resistance. Working on my internship to understand the mechanims of endocytosis of a transporter with a mechanism independent of clahtrins called Glycolipid-lectin mechamism discovered in this institute. I did special immunofluorescence assays to observe this mechanism in an adrenocortical cancer cell line. The project was to assess the control of the metabolism of induced pluripotent stem cells (iPSC) differentiated in Mesenchymal stromal cells through the PD1/PDL1 axis. For that, I: -proceeded to RT-qPCR analysis to identify mRNA coding for metabolic enzymes -performed Seahorse analysis in different conditions to assess the metabolism of this cells by modifying PD1/PDL1 axis with drugs -assessed with FACS (Fluorescence Activated Sorting Cells) the expression of transporter implied in metabolism I performed a small review to sum up the literature context of this project.
