TFM: Development of a DNA vaccine based on replicative RNA.

Director: Dr. Puri Fortes. Co-director: Dr. Pablo Sarobe. Departments: Immunotherapy and Immunology and Gene Therapy. - The aim of this project was to design, produce and test a DNA vaccine platform (NB-PEPI-UM) based on Semliki Forest Virus replicative RNA, containing elements that enhance the 3 signals necessary for the activation of the immune response: a polyepitope (PEPI), an anti-PD-1 nanobody (NB), and Ubiquitin (U) and MHC trafficking (MITD; M) domains to improve antigen processing and presentation. A vector was also produced that would act in trans and encodes the soluble factors XCL1 and FLT3L. I demonstrated nanobody expression by Western blot and PEPI expression by immunizing mice and its antitumor effect in reducing tumor size in a mouse melanoma model and in quantifying IFN-γ produced by immunized mouse spelunocytes. - I performed more than 100 digests, ligation and cloning of plasmids in less than 3 months, also more than 25 Western blots, more than 20 PCRs, 10 ELISAS and ELISPOTs, I also maintained for 8 months several cell lines performing more than 30 transfections with lipofectamine and manipulated more than 100 mice, all this to have as a result an effective vaccine construct and allowed me to gain technical and analytical skills, organization in time managing resources and coordination of different experiments and teams of people. Graduate Ad Honorem Internship in Molecular Pathology of Cancer Navarrabiomed (189 hours)

Tutors: Dr. David Guerrero Setas, clinical biologist of Anatomic Pathology and principal investigator of the Molecular Pathology of Cancer Group and Iñaki Monreal Santesteban, laboratory technician. These practices were dual, had 2 different stays in the Molecular Pathology of Cancer Research Group of Navarrabiomed and the Anatomic Pathology Service of the CHN. Within the internship, besides learning routine work techniques, I was assigned to carry out a small project that consisted of optimizing and fine-tuning the conditions to detect and localize by immunohistochemistry the following markers in paraffin-embedded breast tissue: Claudin 2, carbonic anidrase 9, NMDAR2R, NOX4 and SLAMF9. This work will serve to complement a larger study that will soon be published in an article. The techniques I learned were: bisulfite treatment, pyrosequencing, Western blotting, maintenance of cell lines in in vitro cell cultures, immunohistochemistry, tissue miroarrays, extraction, amplification and quantification of nucleic acids, gene knock out, RT PCR and preparation of library pools for sequencing by Ion Torrent silicon semiconductor chips. FDP: Histological characterization of radiotherapy-induced brain radionecrosis (12 ECTS)

Grade: 9.2 (Outstanding). Tutor: Dr. Diego Serrano Tejero. My FDP in Biology was experimental and I developed it in the Laboratory of Biomarkers and new therapeutic targets for lung cancer led by Dr. Luis Montuenga Badía and belonging to the Solid Tumors Program of CIMA. Techniques learned: immunohistochemistry, double and multiplex immnofluorescence, scanning and bioinformatics image analysis with ImageJ and Aperio and Nissls staining. Summary: Cerebral radionecroses are among the major adverse effects of stereotactic radiotherapy treatments employed to treat brain tumors. Their pathophysiology is thought to be caused by damage to endothelial and glia cells, leading to hemorrhage, hypoxia and brain necrosis accompanied by angiogenesis and inflammation. There are indications that the STAT3 protein regulates these processes and could therefore be an effective therapeutic target to combat radionecrosis. The objectives of the present work are a) To verify whether the drug Eurosil 85 is effective in reducing STAT3 (pSTAT3) phosphorylation levels in brain radionecrosis. b) To set up a multiplex system (using the Vectra Polaris system of Akoya Biosciences) to colocalize and quantify GFAP, F4/80 and pSTAT3 in radionecrosis. c) To histologically characterize radionecrosis lesions. A group of UNAV students formed a multidisciplinary team that carried out the development of a business idea about the implementation of an online marketplace focused on local organic products. We were awarded when we reached the semi-final stage and for me it was an enriching experience because of what I learned and the people I met, helping me to improve my analytical, leadership, teamwork and social skills. Grade: 9 (Outstanding).Supervisor: Dr. Ricardo Ibañez Gastón.This work was developed over 4 months, it represented almost the entire grade of the subject of Plant Biodiversity and required a great effort to do it because it was totally practical and consisted of a taxonomic classification work on a group of plants. In my case I did it on angiosperms and I titled it: Angiosperms of the Pamplona basin and surroundings.I identified 66 different specimens: 50 at species level, 9 at genus level and 7 at family level, with a biodiversity of 29 different families. - Recognized with the award for the best business idea in the biosanitary field.🎖😊 Innovation Factory is the innovation department of the UNAV and develops a competition called Fast Track in which multidisciplinary teams are formed among students to find an idea to improve the world and develop it into a project that can go to market. We received training and feedback from UNAV professors and our work was tutored by Dámaso Molero CEO of 3P Biopharmaceuticals. In the team I proposed the idea we worked on and I played the role of CEO. The business idea we developed consisted in the application of oral probiotics based on Streptococcus dentisani 🧫🔬 bacteria as a viable proposal to prevent and treat caries🦷. In the team we were students from different disciplines from science, to marketing, ADE or engineering so it was important to develop a good communication. We also collaborated with institutions such as SODENA or CEIN who advised us and with the pharmaceutical company AB-Biotics.💊 It helped me with organization and working under pressure since I had to combine it with the third year of my degree in Biology.
