An investigative study on the modulation of vascular tone in mouse aortic vessels (Master’s Thesis). • C57BL/6 mice were dissected to isolate the aortic vessel. • The vessels were then prepared and mounted on the wire myograph. • Dosage Response Curves were generated in response to agonists (and blocked by antagonists) of the Sphingolipid pathway, to study the modulation of the vascular tone in the aorta. Analyzing the anti-angiogenic properties of peptides isolated from shark muscle and visceral mass hydrolysates. (Undergraduate Thesis) • Protein hydrolysates were isolated through enzymatic treatment. • Anti-oxidant (DPPH) and anti-inflammatory (albumin denaturation) assays were used to assess the bioactivity of the hydrolysate samples (due to the underlying connection between oxidation, inflammation and angiogenesis pathways). • The hydrolysates samples with the highest bioactivity were fractionated through Ultrafiltration (3 and 10 kDa filters), the bioactivity of each fraction was then assessed through anti-oxidant and anti-inflammatory assays. • The fractions with the highest bioactivity were purified through Gel Filtration Chromatography (Sephadex G-25). • The anti-angiogenic properties of the purified peptides were assessed through the Chick Chorio-Allantoic Membrane Assay (CAM’s were pre-treated with VEGF). The peptide dosage to be administered to the CAM’s was determined using MTT Assay (MCF-7) Analyzing the anti-oxidant properties of peptides isolated from tuna muscle hydrolysates. • Protein hydrolysates were isolated through enzymatic treatment. • Anti-oxidant assays (DPPH and Reducing Power Assay) were used to assess the anti-oxidant properties of the protein hydrolysates. • Hydrolysate samples showing the highest anti-oxidant properties were fractionated using Ultrafiltration (3 and 10 kDa filters). • Anti-oxidant assays (DPPH and Reducing Power Assay) were used to assess the fraction with the highest anti-oxidant properties.
