Supervisor: Daniel Griffith Anderson, PhD • Developed an IVT-mRNA system for in vivo delivery of the humanized anti-HER2 antibody trastuzumab and demonstrated its anticancer activity in vitro using human breast cancer cell lines, ex vivo using an ADCC assay, and in vivo using a mouse xenograft model. • Developed a protocol for Antibody-Dependent Cell-Mediated Cytotoxicity (ADCC) assay. • Engineered a breast cancer mouse model for analysis of trastuzumab mRNA efficacy. • Performed high-throughput loss-of-function shRNA-based screening. Identified a new lncRNAs (ASTILCS) essential for survival of human liver cancer cells and characterized its molecular mechanism. • Collaborated with the Robert Langer lab, designed and executed in vitro and in vivo experiments to test anticancer activity of novel drug-excipient combinations predicted by a computer-aided drug design. • Performed molecular cloning (Golden Gate, Gibson assembly, Gateway, traditional), cell culture (primary cells and established cell lines), flow cytometry (TUNEL assay, BrdU and Click-iT EdU cell cycle analysis), Western blotting, qPCR, IgG purification, IVT-mRNA synthesis, ELISA, high-throughput genetic screening (RNAi), lentiviral transduction, gene knockdown (using RNAi, CRISPRi, antisense oligonucleotides), cell transformation (Lipofectamine, lentiviral), cell-based assays (survival, proliferation), PCR primer design, sgRNA/shRNA design. Proficient in using SnapGene, ImageJ, FlowJo, ModFit, GraphPad Prism. • Worked with mice: hydrodynamic and tail vein injections, tail vein and submandibular bleeding, generation of subcutaneous xenografts, generation of liver cancer genetic model, survival experiments, dissection • Hired, mentored, and trained technicians, students, and postdocs Show less

Worked on PhD project: • Studied the effects of natural antioxidant carnosine and its derivatives on cell cycle progression in pheochromocytoma cells (PC-12). Performed structure–function analysis to investigate the mechanism of carnosine antiproliferative effect. • Characterized carnosine’s protective effects on cerebellar neurons oxidative damage. • In collaboration with St.-Petersburg Institute of Bioregulation and Gerontology described the effects of synthetic antioxidant pinealon on cancer cell survival and cell cycle progression. • In collaboration with the Department of Biochemistry of Russian Peoples' Friendship University in Moscow investigated molecular mechanism of antitumor effect of L-Amino acid oxidases. • Performed: flow cytometry (cell cycle analysis with PI, ROS analysis with DCFH-DA), cell culture (PC-12), immunoblotting, qRT-PCR.
