Supervisor: Daniel Griffith Anderson, PhD • Developed an IVT-mRNA system for in vivo delivery of the humanized anti-HER2 antibody trastuzumab and demonstrated its anticancer activity in vitro using human breast cancer cell lines, ex vivo using an ADCC assay, and in vivo using a mouse xenograft model. • Developed a protocol for Antibody-Dependent Cell-Mediated Cytotoxicity (ADCC) assay. • Engineered a breast cancer mouse model for analysis of trastuzumab mRNA efficacy. • Performed high-throughput loss-of-function shRNA-based screening. Identified a new lncRNAs (ASTILCS) essential for survival of human liver cancer cells and characterized its molecular mechanism. • Collaborated with the Robert Langer lab, designed and executed in vitro and in vivo experiments to test anticancer activity of novel drug-excipient combinations predicted by a computer-aided drug design. • Performed molecular cloning (Golden Gate, Gibson assembly, Gateway, traditional), cell culture (primary cells and established cell lines), flow cytometry (TUNEL assay, BrdU and Click-iT EdU cell cycle analysis), Western blotting, qPCR, IgG purification, IVT-mRNA synthesis, ELISA, high-throughput genetic screening (RNAi), lentiviral transduction, gene knockdown (using RNAi, CRISPRi, antisense oligonucleotides), cell transformation (Lipofectamine, lentiviral), cell-based assays (survival, proliferation), PCR primer design, sgRNA/shRNA design.
