I studied the the biochemistry of human NEET family proteins, Iron-Sulfur proteins, right now. mitoNEET, an Outer Mitochondria Membrane associated protein, is the binding target of the type 2 diabetes medication pioglitazone, which helps to reduce the insulin resistance, and improve the metabolism of the patient. Over-expression of mitoNEET is also observed in various types of cancer cells. My study illustrated the role of mitoNEET as a central hub of a de novo Electron Transduction Pathway on the mitochondria, that may play a key role in cancer or diabetes related metabolism, and be regulated by the medication pioglitazone. I worked on the molecular mechanism of interaction between Miner2, a Mitochondria matrix located human NEET protein, Iron-Sulfur cluster and Ntric Oxide (NO), a key molecular regulated various biological functions, like Vasodilation, Neurotransmission, Cell killing by immune cell, etc as well. During the internship in Shanghai Institute of Immunology, I am trained to study the molecular mechanism of key signaling transduction pathway (mTOR2 pathway), which is one of the universal regulation mechanisms for all kinds of immune cells. My training focused on the cellular study. I am familiar with the working pipeline in a typical immunology lab. I shadowed senior research scientist throughout each step of research projects, and practiced some of them myself, like harvesting immune cell from mice with flow cytometer, expressing external protein in cultured cell lines (Hela, 293T, MEF), Knocking out/down gene expression with RNA tools, constructing fusion protein with fluorescent tag to study their cellular localization and interaction with other kinds of protein by fluorescent or confocal microscopy. Besides that, I also spent time and effort on basic molecular laboratory skills training, like PCR, Plasmid Construction, and Western Blot, etc. Show less
