I studied a plant protein toxin, ricin, on its interaction with its cellular substrate, the ribosome. - Identified Arg235 to be the most critical amino acid for electrostatic interactions between ricin A chain and the ribosome P stalk. (Scientific Reports) - Identified a hydrophobic pocket near Arg235 to be important for ribosomal interactions and conducted mutagenesis studies systematically on functions of critical residues. (Bioscience Reports) - Performed a detailed review on in… Show more I studied a plant protein toxin, ricin, on its interaction with its cellular substrate, the ribosome. (Bioscience Reports) - Performed a detailed review on in vitro and in vivo enzymatic assays critical for ribosome-inactivating protein functional studies. (Toxins) - Developed assays that successfully identified ribosomal protein genes critical for toxicity of ricin from a screening of yeast ribosomal protein gene deletion library. - Presented results at national conferences (2015 Proteins Gordon Research Conference and 2014 Translational Control Cold Spring Harbor Conference). - Mentored 5 undergraduate students. Lead to continued employment of one student at Rutgers University, another student presented poster in the research-intensive summer experience (RiSE) program and at the 2014 Annual Biomedical Research Conference for Minority Students. Technique keywords: Cloning, vector design, mutagenesis. Recombinant protein expression (high-throughput protein expression condition screening) and purification (AKTA, affinity chromatography, SEC, IEX, heparin etc.) Enzyme characterization, e.g Michaelis-Menten kinetics, enzymatic activity tests. Protein structural visualization (UCSF Chimera, Pymol), figure generation and movie making. Protein-protein interaction, surface plasmon resonance, on GE Biacore system. Real-time PCR (qPCR), RNA based assays. Yeast culture, transformation, viability tests, RNA isolation, etc. Mammalian transfection/EGFP cotransfection, viability test, protein translation inhibition tests, etc. ELISA, flow cytometry, SDS-PAGE, western blot.
