I have had the pleasure of working with Wessel, a bright young scientist and Software Engineer, who would be a valuable asset for any company. Wessel is a great contributor who consistently completes tasks assigned to him in a timely manner, and is capable of efficiently handling multiple projects simultaneously. Global Factories is a producer of machines that repack and verify different types of medicine into cup and blister cards or pouches to improve adherence and safety of distribution of medicines and lower the risk of re-admittance of patients to hospitals. These machines produce enormous amount of data that can be used to monitor and improve their performance. As a Data Scientist my tasks include: - Maintaining our Microsoft Azure cloud environment, including Data Lake storages that contain telemetry data that is sent by machines that are located at customers and distributors of Global Factories. - Extract, transform and load (ETL) big data (JSON files) with Apache Spark (Python/Scala) using Azure Synapse Analytics. - Creating SQL serverless views on the Delta Lake in Azure Synapse Analytics. - Visualise and analyse data in Microsoft PowerBI for several departments, such as software development, sales, purchase and customer care. - Feature engineering and building Machine Learning algorithms (e.g. decision tree classifiers, lineair regression) to analyse data. - Developing Azure Functions (C#) to extract or process data. Besides, I followed a C# software development course at BetaBit. This program covered: C# fundamentals, object-oriënted programming, interfaces, generics, working with nulls, defensive coding, error handling with exceptions, asynchronous programming, unit and mock testing, dependency injection and LINQ. Photolyases can repair UV-induced DNA damages by photo-reactivation using visible light as energy source. We believed that photolyases can be used as an effective tool to investigate damage specific binding and dissociation kinetics of nucleotide excision repair proteins. My work therefore extended the use of photolyases to study NER proteins by using live-cell imaging. I have made an attempt to elucidate the mechanism of ABCC6-mediated ATP release with bioluminescence-based assays and a relatively new protein identification method, BioID. In addition, I tried to determine the functionality of ABCC6 expressed in yeast. Graduation project / Scientific research to Primary Ciliary Dyskinesia (PCD). Primary ciliary dyskinesia (PCD) is an autosomal recessive disorder characterized by impaired ciliary function in the upper- and lower respiratory tract. Patients suffer from multiple respiratory tract infections, eventually leading to permanent lung damage (bronchiectasis). It is a rare condition, which occurs in 1: 15.000-30.000 newborns. An early diagnosis of PCD is essential, so treatment to preserve lung function can start as soon as possible. However, the diagnosis of PCD in children is difficult as there is no gold standard test. The aim of this project was to test a targeted-exome sequencing approach for the diagnosis of patients with PCD. Techniques used: - DNA isolation - RNA isolation - PCR - Agarose gel electrophoresis - Sanger sequencing - Library preparation for targeted sequencing (Illumina) - Library hybridization with targeted kit (illumina) - Next-generation sequencing (HiSeq2500) - Software used: Integrative Genomics Viewer v2.3.5 and Alamut v2.4 Show less
