• Thesis title: Studies on mitochondrial J-domain proteins and J-like proteins in Arabidopsis thaliana using CRISPR and in silico approaches• Project description: Worked for investigating the biological significance of mitochondrial J-domain proteins (JDPs) in Arabidopsis thaliana using the CRISPR-Cas9 mediated gene knockdown approach. Also, employed Saccharomyces cerevisiae as a model system to better understand the functioning of the JDP:JLP apparatus.• Developed CRISPR/Cas9 mediated knockout lines for all the three Pam18 (a mitochondrial J-domain protein) orthologues in Arabidopsis thaliana and screened using fluorescent molecular markers• Modelled A. thaliana J-like proteins (JLPs) and performed molecular docking studies with J-domain proteins (JDPs) to determine their evolutionary relationship with putative interactors• Studied the effect of overexpression of A. thaliana JLPs on the phenotype of heterologous systems like Saccharomyces cerevisiae• Learned molecular cloning, transformation, CRISPR/Cas9 KO, phenotyping, genotyping, confocal microscopy, qRT-PCR, conventional PCR variants, protein expression, western blot• Project Supervisor: Dr. Chandan Sahi (Associate Professor and Department Head, Biological Sciences, IISER Bhopal)
