Title of the MSc Thesis: “Αdhesion and biofilm formation capacity of the wine spoilage yeast, Brettanomyces bruxellensis” -Participation in the design of the experiments -Traditional cultural microbiological methods -Wine sensory analysis -Fourier-transform infrared spectroscopy (FTIR) The main object of the study was to evaluate the adhesion and biofilm ability of the wine spoilage yeast Brettanomyces bruxellensis. Adhesion ability of eleven strains was initially studied and at the same time, the metabolic fingerprint of planktonic versus adherent cells was compared using FTIR. A strain with a high spoilage potential was then selected for the in-depth study of biofilm formation. Specifically, the factors that affect adhesion, the biofilm kinetics and the possible spoilage of wine by attached cells were examined. For this purpose, different conditions (low pH, ethanol, sulfur dioxide, wine, must) and materials (stainless steel, glass) found in winemaking were evaluated as factors affecting adhesion. Stainless steel coupons were used for the biofilm growth over time and sampling took place at 0, 24, 48, 72 and 144 hours, while for the spoilage potential the "contaminated" red wine was incubated at 25oC for two months. Finally, new techniques were used to treat the adhesion of B. bruxellensis (33.1) to stainless steel coupons, such as the treatment with different concentrations of chitosan and the presence of two different commercial strains of the lactic acid bacteria Oenococcus oeni. Additional experiments: The factors that affect adhesion, the biofilm kinetics and the possible spoilage of wine by attached cells were examined also for Saccharomyces cerevisiae and Schizosaccharomyces pombe.       I had three months of my internship done in the Disease Systems Lab (Animal & Veterinary Sciences Research Group) of SRUC (Scotland’s Rural College) where I investigated the literature and prepared a written report on the role of pregnancy hormones on immunity to pathogens which I presented to a scientific audience at the department. After shadowing a PhD student, I performed laboratory techniques such as RNA extraction, cDNA synthesis, standard PCR and real time quantitative PCR. I also helped in animal experiments (mice dissections and monitoring of data during post mortems and sheep dissections and sample collection during post mortems) and received training on how to design primers.
