Provided pathophysiology and scientific and technical support for research, development and manufacturing of a broad spectrum of healthcare products including medical devices and small molecule pharmaceutical compounds. Provided guidance and support for the design, development and implementation of research and development studies and laboratory procedures to ensure GLP compliance and establish technical requirements. Provided histopathology support: several 30 day toxicity studies on extractables and leachables (including peer review); toxicity assessment of a common leachable and on a potential impurity; model optimization for four animal models of human disease; safety assessment of a product from a new supplier; and immunohistochemistry evaluations. Performed several large literature database searches for nonclinical safety assessment white papers, assisted in a PBRER evaluation of a marketed product and performed due diligence to evaluate the benefit and scientific soundness of several research proposals for external funding. Managed a team of 4 technicians and the histology laboratory Contributed and led cross-functional management teams aimed at continuous improvement and audit readiness Corrective Action, Preventive Action (CAPA) training Show less

Managed studies for continuation of thesis project. Performed required experiments (see below). Performed peer review of scientific documents Graduate thesis work focused on Mycobacterium tuberculosis and innate immunity within the lung with a specific focus on the innate immune defense molecule, Surfactant Protein D (SP-D), which showed in vitro host protective effects. The goal was to define the mechanism of and enhance the protective effects of SP-D in the natural history of tuberculosis infection. Cell culture: primary (human and murine) and cell line (THP-1, CHO) cell culture and maintenance Immunological Assays: ELISA, Western blots, confocal, flow cytometry +/- cell sorting, (co)immunoprecipitations, lectin immunoblots (SP-D) Microbiology: bacterial culture of Mycobacterial species, Rhodococcus equi, Escherichia coli; cell wall molecule isolation protocols specific for Corynebacteriaceae; fluorescence microscopy for cell association assays Trained to work in a Biosafety Level-2 and Biosafety Level-3 Biochemistry: Protein isolation from eukaryotic cells – size exclusion and saccharide affinity chromatography, dialysis, protein sequencing Molecular biology: DNA (plasmid) isolation, site-directed mutagenesis, PCR, transformation and transfection (including siRNA knockdown) Animal studies: in vivo infection experiments; Maintenance of a knockout colony Show less
