Cancer is a multistep process that involves mutations and other alterations in oncogenes and tumor suppressor genes. Genome sequencing studies have identified a large collection of genetic alterations that occur in human cancers. However, the determination of which mutations are causally related to tumorigenesis remains a major challenge. The goal of our lab is to create and utilize genetically engineered mouse models (GEMMs) to model various types of cancers with a particular focus on lung cancer. These models enable the characterization of the critical steps of tumor initiation and progression at a molecular, cellular and whole organism level. Our lab is pioneering the use of GEMMs of lung cancer in combination with in vivo CRISPR/Cas9 genome editing approaches to address the immediate need to systematically interrogate the complex catalog of mutations obtained from cancer genome sequencing studies.
