Daejeon, South Korea I am currently working on generating SCON (Short Conditional intrON) knock-in within the SOX2 gene of zebrafish using CRISPR-Cas9. This novel genetic construct behaves like an intron in its uninduced state but can prematurely terminate translation, specifically upon cre-based induction. Hence allowing us to generate scarless conditional knockout alleles in vivo. This approach could be very useful in studying developmentally important gene functions in the adult stage.

Pilani, Rajasthan, India • Worked under Prof. Syamantak Majumder on RNA methylation in TGF-Beta induced endothelial to mesenchymal transition. Our findings demonstrated that the induction of TGF-Beta indeed resulted in EMT, characterized by cellular changes. We observed no significant alterations in the overall RNA methylation levels. Additionally, the expression of the methyltransferase complex remained unchanged. • Worked under Prof. Meghana Tare on neurodegenerative diseases - performed climbing and lifespan assay on the Drosophila model of Parkinson's and Alzheimer's. Parkin RNAi males showed a 50% decline in climbing ability. Tau flies exhibited a gradual reduction in climbing ability, but the trend was not statistically significant. I also worked on a CRISPR strategy to create drosophila mutant model.

Pune, Maharashtra, India I standardized flight assay for the lab, wherein I was able to show that, wing defect in Drosophila P58S start to appear from the 3rd day onwards, learnt in-silico design of ssODN vectors to make point mutation, learnt DAM assay and rhythmically to analyse the DAM assay data.
