My primary focus is on tobacco, obesity, physicl activity and nutrition, diabetes, and heart disease and stroke; however, I have been cross training in Maternal and Child Health as well. I construct theories on risk factor association with chronic diseases, and develop and distribute reports statewide to stakeholders and health department staff. After completion of my Master`s in Public Health (MPH), I was awarded an Emerging Infectious Diseases (EID) Fellowship through the Centers for Disease Control and the Association of Public Health Laboratories. As an EID fellow I learned a variety of techniques within a clinical lab setting at the Colorado Department of Public Health and Environment. I did a rotation through the Serology, Public Health Microbiology, Molecular Science, and Environmental Microbiology laboratories learning a variety of techniques including but not limited to: MagnaPure and Bio-Mek extraction; enzyme immunoassays (EIA) for HIV, Hepatitis B and C, and Hantavirus; Salmonella sp. and Escherichia coli serotyping and biochemical testing techniques used for typing; routine diagnostic testing using real-time PCR for pertussis, plague, tularemia, and norovirus using the SmartCycler II platform; implementation and validation of a multiplexing immunoassay using the Bio-Plex platform for Salmonella serotyping; and cystic fibrosis (CF) luminex assay using the Bio-plex platform for determination of specific mutations from dry blood spots for newborn screening. My most significant accomplishments as a fellow were: the validation and implementation of a quantitative reverse-transcriptase (q-RT) PCR for Rabies specimens, sequencing of Rabies specimens using the Beckman Coulter CEG 8000 sequencer, routine Shiga-toxin (stx) real-time PCR on O157 and non-O157 Shiga-Toxin producing E. coli (STEC) specimens and organization of data. As a laboratory technician during my graduate coursework, I was able to apply a number of microbiology techniques I had learned during my undergraduate coursework in a research laboratory setting. While researching iron transport systems, manganese transport systems, and biofilm formation of Yersinia pestis, I had the opportunity to work on a number of experiments requiring different microbiology techniques. One such technique was the crystal violet assay, which was used to quantitatively determine the amount of biofilm produced by the bacteria. I also used such methods as Western Blot Analysis using SDS-PAGE gel purification to determine protein levels, Southern Blot Analysis (hybridization) using a probe to detect DNA, phage induction which allowed for integration of the phage into the host_s genome, and polymerase chain reaction (PCR) using taq, proofstart, and pfu polymerases to perform cloning of genes, genotyping of specific mutations, random mutagenesis, site-directed mutagenesis, and red-recombinase mutagenesis. I also used Beta-galactosidase assays to determine promoter activity for promoters which have been fused to the lacZ gene. Researching with these techniques has also allowed me to design my own primers, work with various restriction enzymes, perform plasmid preparations using a number of methods, LMA gel extractions, gel electrophoresis, and sequencing mutations using DNAMAN. Developed skills for tabulating and summarizing studies. Compiled data from charts gathered during a study into an excel spreadsheet and analyzed a number of variables, determining the distribution for certain variables and establishing appropriate cutoffs for analysis. Computed z scores on the cities of Pan de Vida and Juan XIII for underweight, overweight, wasting, and stunting using WHO Global Database on Child Growth and Malnutrition tables. Calculated means and percentages for variables of particular interest for future study. Categorized certain variables to better organize the data to determine what the expectations may be for a similar study to be conducted the following year. Organized literature on Arsenic reproductive outcomes and condensed the information for a poster presentation. Entered data into Microsoft Access. Initiated a literature review and defined a question for my Capstone (Thesis). Information on human papillomavirus and cervical cancer knowledge in two Appalachian counties in Kentucky gathered during a telephone survey. Familiarized myself with using the statistical program STATA. Learned to recode variables and conduct Pearson chi-square p values and multiple logistic regression analysis. Show less
