I am currently investigating the molecular mechanism of how the Erythropoietin-producing Human Hepatocellular Receptors (Eph) family and their ligands (Ephrins) interact in trans and in cis on cell membranes using traditional in vitro, ex vivo, and in vivo biophysical, biochemical, and biological assays. I am also investigating how the different Eph-ephrin expressions on cancer cells and/or hosts affect host development, immunosurveillance, tumorigenesis, and metastasis. Routinely used techniques include culturing model and primary cancer cell line, immunoblotting, modifying the genetic profile of the cell lines using retro/lentiviral expression system, CRISPR/Cas9 knockout system, profiling RNA level expression of proteins real-time polymerase chain reaction, profiling immune components using flow cytometry, FACS sorting, fluorescence-based imaging methods, and performing quantitative and qualitative data analysis. I studied transmembrane protein assemblies under the presence/absence of their ligands, specifically by: - quantitatively assessing basal homotypic and heterotypic interactions of the Human Epidermal Growth Factor Receptor (HER) family, - identifying their homotypic and heterotypic interaction interfaces, and understand the effects of their oncogenic mutations on such interactions using PIE-FCCS (Pulsed interleaved excitation-Fluorescence cross-correlation spectroscopy),- identifying the interaction interface of Erythropoietin-producing Human Hepatocellular Receptors (Eph) family and their ligands,- understanding pan-RTK interfamily interaction.I also studied the effects of peptides on EphA2 receptor assembly for potential drug development and copper-induced clustering of phosphatidylserine lipids with model lipid membrane interface.
