Advisor: Caroline Ajo-Franklin In Ajo-Franklin's lab, I am focusing on characterizing and engineering the extracellular electron transfer pathway in probiotic lactic acid bacteria and in microbial community. The long term goal of my project is to creat probiotic bioelectronic sensors to assist rapid drug monitoring and disease diagnosis in situ. Advisors: Ke Lan & Shuwen WuUndergraduate thesis: The interaction between viral 3D polymerase and host cells during EV71 infectionScreen interacted proteins with Enterovirus 71 3D polymerase by IP-MS• Overexpressed 3D-Flag fusion protein in 293T under the infection background of EV71 & immunoprecipitated the 3D-Flag protein by anti-flag beads• Proteomics analysis and dissected the protein interaction patterns• Studied the interaction between ANXA2 and 3D polymerase with cell biology methods assisted by co-IP, CRISPR-Cas9, lentivirus integration, and immunofluorescence. High-throughput screens and studies of Enterovirus 71 sensitivity and resistance to inhibitors• Screened four thousand drugs & discovered four potent drugs against EV71• Performed time-course experiments to understand the mechanism of action of the drugs• Measured EC50 and CC50 of the drugs by qPCR and MTT assay, respectively• Screened drug-resistant strains of EV71 & performed whole-genome sequencing to find the mutated sites• Conducted mice experiments to test the drug potency in vivo Show less
