Investigated changes in brain development in mice knocked out for dyslexia susceptibility gene DCDC2. -Preformed whole cell patch clamp experiments on layer IV primary auditory cortex pyramidal neurons in acute slice preparations from P15-P18 mice. -Cloned ChETA, eNpHR3.0, and GCAMP3 genes into plasmid vectors for use with in utero electroporation and piggyBAC transposon gene delivery techniques.Designed a project examining relationship of columnar circuit activity and local astrocyte activity. -Combined delivery of GCAMP3, ChETA, and piggyBAC plasmids to neural progenitor tissue with in utero electroporation technique in rats. -Prepared acute slices and investigated relationship using two-photon microscopy, laser/fiber-optic stimulation, and image analysis. Used image analysis software including Adobe Photoshop, imageJ, and Neurolucida. Research culminated with an undergraduate thesis on combining the use of the piggyBac transposon and in utero electroporation as a novel technique for stable transgene expression, thus completely labeling cell lineage with cell-type specific fluorescent tags.
