Nagaraja Generation of Rv3788 mutants through Overlapping PCR and molecular cloning:Rv3788 is a Gre protein homologue in Mycobacterium tuberculosis found to bind the secondary channel of RNAP and inhibit transcription. In this project, I created site-directed mutations using overlapping PCR in the amino acid sequence of Rv3788, expressed it in E.coli and purified the protein. The resulting changes in mutant protein’s activities provided necessary insight into the mechanism of Rv3788’s inhibitory function. Show less

      Validation of pab1 as a bonafide molecule responsible for cytokinesis checkpoint maintenance: with Dr. Mithilesh Mishra In Schizosaccharomyces pombe, the phosphatase Clp-1 encompasses a cytokinesis surveillance mechanism which is essential upon any cytokinetic challenge to the cell. The gene pab-1 was suspected to be a downstream effector of Clp-1. To validate this, pab-1’s essentiality in a background of artificially created damage to the cytokinetic ring was studied. Double mutants of pab-1 deletion and various temperature sensitive ring components were generated and checked for viability and phenotype. Show less
