During the project, I have looked at how neutrophils engulf different sized bacteria and the importance of PI3K and actin during the phagocytosis process. I accomplished this by taking blood samples from healthy donors, isolating the neutrophils form the samples and treating them with different sized latex beads. Then, PI3K and actin were inhibited to compare the results to non-inhibited samples. I have found that PI3K and actin are crucial for phagocytosis of larger particles. Neutrophils were able to engulf smaller particles even when PI3K was inhibited. I observed and counted the cells under fluorescence microscopy and performed statistical analysis to write my finishing thesis. In addition to the exceptional laboratory skills, that I gained during this project, I have also learnt how to organise my time in the lab to produce high quality results in a limited time frame. The prion disease is caused by missfolded proteins that forms plaques in the brain and result in neurodegenerative disorders. There is currently no cure for this disease and chances of an outbreak is relatively high as cattles can transmit these proteins to humans. During my time in the lab I have studied the chemical behaviour of these proteins when they interact with different metals. Firstly I grew genetically… Show more I have taken part in a research project on prions at Dr. Fiona Houston's lab. The prion disease is caused by missfolded proteins that forms plaques in the brain and result in neurodegenerative disorders. There is currently no cure for this disease and chances of an outbreak is relatively high as cattles can transmit these proteins to humans. During my time in the lab I have studied the chemical behaviour of these proteins when they interact with different metals. Firstly I grew genetically modified bacteria in large scale, that can produce these proteins. Afterwards I purified these proteins by using fast protein liquid chromatography and treated them with different metals. These were then tested by running SDS and LDS-page gels. By increasing the understanding of the molecular behaviour of these proteins, we can aim to develop a treatment that can block the uncontrollable growth of prions in the brain. I also performed RT-QuIC analysis on these samples in BSL-3 level lab. The data that I have collected will be published in the paper that a research fellow, Charlotte Thomas, is working on now.
