Optimized several highthroughput cell-based assays using vesicular stomatitis virus (VSV). Titered VSV using plaque assay. Tested for presence of EBOV and MARV proteins of mouse sera and antibody samples from collaborator's lab using Western Blot. Performed mycoplasma detection assays using EnVision Plate Reader. Transfected HEK293T cells using Lipofectamine 2000. Used Lipofectamine RNAiMAX to knockdown genes in HEK293T and A549 cells. Used EBOV minigenome highthroughput screening system to assess activity and cell cytotoxicity of multiple compounds. Optimized VSV antiviral assay to screen activity and cell cytotoxicity of compounds. Quantified fluorescence units of VSV-GFP and Influenza A/PR8-GFP using EnVision and BioTek Cytation 5. Visualized VSV-GFP and PR8-GFP-infected cells using EVoS Digital Microscope. Isolated VSV and Zika-infected RNA samples using TRIzol and generated cDNA using SuperScript III First-Strand Synthesis System for using qrt-PCR to detect levels of viral synthesis. Measured antiviral activity of compounds using ZIKV MR-766. Measured OD of bacterial strains. Performed resazurin microtiter assay for determining drug resistance in M. smegmatis. Experienced in basic mouse handling techniques such as anesthesizing mice using isoflurane and performing euthanasia by cardiac puncture, administering CO2 and cervical dislocation. Utilized mouse models for isolating bone-marrow derived macrophages and measuring antibody levels by intraperitoneal injection of VSV-GP and VSV-GFP.
