The main aim of my PhD project was characterizing the role of BCAP (B-cell adapter for PI3K), which has been shown to associate with and regulate the function of PI3K resulting in negative regulation of inflammation induced by TLR activation. In my PhD project, I mastered several techniques:-Protein expression and purification to solve the 3D structure of the N-terminus domain of BCAP, which confirmed its role in innate immunity. -Physical Biochemistry characterisation of BCAP.-Studying the protein-protein interaction between BCAP and PI3K by expressing both full-length proteins by Baculovirus system.-Pull-down assays to confirm the interaction between BCAP and PI3K, TLR, and PLCγ2.-Yeast two-hybrid assay and screen.
