Dr. Karl M Thompson, Ph.D.

I worked with Sai in the Lewis Laboratory at the Philips Institute for Oral and Craniofacial Molecular Biology for a little over a year. At the time, I was a postdoctoral research fellow and Sai was a research / laboratory technician. Sai was extremely helpful to everyone within the laboratory. Unofficially, she acted as a laboratory manager as well as a research technician and her organizational skills and expertise were invaluable for the execution of all of the lab projects. Sai will be an excellent asset to any laboratory or organization that she becomes a part of.

Cheyanne Warren

Sai was an extremely dedicated laboratory research specialist in our lab. Her knowledge of research techniques and her attention to detail made her a valuable member of the lab. She also has a wonderful disposition which makes easy to work with!

      • Elucidation of the mechanisms of Iron acquisition and adhesion of oral anaerobic bacteria; Porphyromonas gingivalis and Prevotella intermedia.• Determination of the contribution of the proteins mediating Iron uptake or adhesion to host cells to virulence of the organism.• Developing different mutants deficient in Iron uptake proteins and characterizing their ability for the uptake and transport of Iron.• Mutants deficient in Iron dependent regulators are examined for their ability to regulate the genes using Microarrays, Chromatin Immunoprecipitation and DNA binding assays using the recombinant form of the purified proteins.• Detection and identification of protein forming complexes that helps in hemin transport in P. gingivalis that involves Immunoprecipitation of proteins binding to known hemin transport proteins like HmuR and HmuY and identification of the complexes by mass spectrometry.• Inactivation of the genes encoding potential adhesins and determination of the properties of mutant strains and the recombinant proteins in their ability to bind to host components. • Perform assays using bacterial cultures, compile data, and assist other members with their projects.• Maintaining cultures of the bacterial cells, developing allelic exchange mutants and their characterization, preparation and characterization of recombinant proteins. • To extract RNA and proteins, perform standard blotting techniques for the analysis of recombinant proteins and nucleic acids, perform microarray analysis and interpret the data. • Responsible for project ownership for safety, lab equipment maintenance, management of troubleshooting activities, regular monitoring of lab equipment and project ownership which involves research, planning, infrastructure development, defect reduction.• Developing new processes, recipes, and optimize protocols.• Acted as a trainer and mentor to Pre-med students, Pre-dental students, Undergrad students, Master’s students and technicians.

Project: Determination of Erythrocyte Binding Specificity of MSP7• The objective is to determine the erythrocyte binding specificity of MSP7 (Merozoite Surface Protein).• The work involves cloning of MSP7 into PRE4 and transfecting into COS7 cells.• Identify the cells that are expressed and observe if the same are expressed in human RBC also.• This is achieved by cloning MSP7 into PRE4 that allows the expression of heterologous protein on the surface of mammalian cells. These cells are then incubated with human erythrocytes.

    • Volunteer Research Assistant, Department of Human Genetics
      • Aug 2006 - Jan 2007

      • Had an opportunity to volunteer a research lab rotation in the Department of Human Genetics, Virginia Commonwealth University. My work was mainly focused on Treacher Collins Syndrome. • Implemented Recombinant DNA techniques such as designing primers, PCR, site directed mutagenesis, Restriction enzyme analysis, Agarose gel electrophoresis, SDS PAGE, Plasmid preparation and purification.
