Human genetics research aimed at expanding our knowledge on how certain inherited genetic variants can contribute to the development of a broad range of cancers. Functional analysis of germline variants in DNA repair genes, specifically genes involved in double strand break repair and nucleotide excision repair. Use of cellular model systems and assays to examine the biological consequences genetic variants on protein function and cellular pathways as well as xenograft models to determine drug responses in certain mutational backgrounds. o Identification and evaluation of genetic biomarkers of drug response, preclinical proof-of-concept studies o CRISPR/Cas9 genome editing, specifically precise editing of single nucleotidechanges with application of cutting edge variant detection methods (ddPCR, CRISPR‐seq)o Induced pluripotent stem cell (iPSC) culture, transfection, genome editing, differentiation of iPSCs and re‐programming of patient fibroblast cells into iPSCs Show less

The focus of my work in this role was to identify genetic biomarkers predictive of drug response and to define which patients would benefit most from such targeted therapeutic interventions. To this end, we leveraged information from large next-generation sequencing datasets identifying genetic alterations associated with cancer risk and used CRISPR/Cas9 technology to precisely model these changes. Using these models we were able to analyze the functional consequences of these genetic variants in vitro and in vivo. Show less
