      The research focus of the lab was HIV/AIDS and developing antagonists of the interaction between HIV-1 envelope protein gp120 and its receptors, CD4 and CCR5. A second research focus was asthma and related inflammatory processes. I was responsible for the operation of the protein core, which entailed recombinant protein expression. To this end I utilized several hollow fiber bioreactors for growth of recombinant mammalian and insect cells, as well as roller bottles and shaker flasks. I was responsible for process development and downstream protein purification by chromatography (FPLC) and validation by ELISA, SDS-PAGE, Western blot and SPR analysis. I was responsible for the analysis of serum from guinea pigs immunized with HIV-1 envelope protein mixed with antagonists. For this I developed several direct binding and inhibition ELISA assays. I instructed and supervised students and personnel.

      The main focus of the work was production (fusion) and cloning of hybridomas and production, purification (FPLC) and subsequent testing of monoclonal antibodies, as well as culturing different types of cells as a service. I did EBV-transformation of human lymphocytes and worked with mouse embryonic stem cells. I was using hollow fiber bioreactors and a minibioreactor to optimize conditions for maximum antibody production. In addition I helped setting up the DNA Sequencing Core Facility and running an automated DNA sequencing service. The work included assembly of sequences, homology searches, Blast searches, ORF analysis and general advice. In both facilities I was responsible for the supervision and training of personnel.
