      During this internship at ORCA Therapeutics, situated at the group of prof. dr. V. van Beusechem in the Cancer Center Amsterdam (VUmc), I have investigated a potential improvement of the potent oncolytic adenovirus ORCA-010 (currently being prepared for clinical testing). I have successfully created a novel virus by homologous recombination and assessed its antitumor efficacy on multiple cancer cell lines using in vitro cytotoxicity assays. The findings were presented to and discussed with the group of prof. dr. V. van Beusechem and the staff of ORCA Therapeutics as well as documented in a written report. Show less

      My research project at the department of Biochemistry and Cell Biology focused on the role of posttranslational modifications (PTMs) of the invariant chain (Ii) in the processing and transport of MHC class II in mouse dendritic cell line D1. I have optimized a substitution chemistry protocol for the detection of PTMs and confirmed PTM of Ii using SDS-PAGE and western blotting. In addition, I have created a D1 dendritic cell line in which expression of Ii was silenced by stable transfection with shRNA's. Mutant Ii incapable of acquiring the PTM of interest was reintroduced into these cells to evaluate the role of Ii PTMs using flow cytometry and fluorescence microscopy. Findings were documented in a written report and presented to the research group of prof. dr. W. Stoorvogel. Show less
