      ECNP is an independent scientific association committed to ensuring that advances in the understanding of brain function and human behaviour are translated into better treatments and enhanced public health. In the ECNP Office, I am responsible for overseeing the science-related functions.       First steps towards a Cell Therapy for Myotonic Dystrophy: the myogenic potential of skeletal muscle-derived pericytes from patients.I have always enthusiastically shared my research ideas and results. Subsequently, the once small one-person PhD project nowadays involves numerous clinicians, researchers, patient organizations and students. Since we’ve obtained a substantial grant in 2020, I participate as a post-doc on this project together with five new muscle researchers. I like to involve talented professionals and I can clearly see that a broadly oriented and collaborative attitude brings innovation and progress. During my PhD I have successfully isolated a distinct class of myogenic progenitors, called pericytes, from patients with a muscular dystrophy called myotonic dystrophy type 1 (DM1). The ultimate goal is to develop a systemic cellular therapy to combat muscle atrophy present in this disorder. When I started my PhD project in 2016, I was determined to perform the first pre-clinical trial. With the additional grant and my persistent attitude, I have recently finished this promising work.       PhD project: Development of an Autologous Myogenic Cell Therapy against the Neuromuscular Phenotype of Myotonic Dystrophy type 1 (DM1).We are working towards autologous and CRISPR/Cas-corrected cells that can systemically be applied, renew the progenitor cell pool and fuse with existing myotubes to form regenerating fibers, hereby relieving muscle symptoms in patients with the devastating disease - myotonic dystrophy type 1. Collaborations (stays abroad):- KU Leuven - Stem Cell and Developmental Biology Professor Maurilio Sampaolesi July - August 2016The publications of Professor Sampaolesi inspired us and contributed to the establishment of my PhD project. More detailed knowledge on understanding degenerative and spontaneous regenerative processes in muscles, approaches to promote long-term regeneration and, most importantly, hands-on experience on the purification of progenitors from skeletal muscle were obtained in this group. - University of Glasgow - Institute of Molecular Cell and Systems Biology Professor Darren Monckton August 2019 To determine the potential of pericytes as a source of replacement cells in myotonic dystrophy and to determine the utility of in vitro gene editing, it is important that we are able to precisely measure the CTG repeat length. The unique research techniques of Darren Monckton and Sarah Cumming were indispensable for the progress of this project. By using the SP-PCR technique in Glasgow, we were able to improve our understanding of the behaviour of the repeat over time and in different tissues obtained from our DM1 patient cohort. Show less

Identify nuclear mislocalized microRNAs (and characterize the effect of mislocalization) by performing RNA-seq to compare the nuclear and cytoplasmic distribution of microRNAs and other ncRNAs in both controls and mTLE patients.
