The aim of this internship is to characterize the sensitivity and the dynamic of jREX, novel calcium reporter, in comparison with GCaMP6f, an established calcium reporter that emits green wavelength. Together with phluorin, vesicle exocytosis reporter, we are able to see the vesicle dynamics in relation to the calcium level. This study is performed in hippocampal primary neurons using live-cell imaging and patch-clamp under-supervision of Roberta Mancini and Alexander J.A. Groffen. We used patch-clamp, optogenetics, and several other molecular approaches to investigate the corelease of Acetylcholine (ACh) and GABA in layer I and VI medial prefrontal cortex (mPFC). This study was conducted in department of Integrative neurophysiology under supervision of Tim Heistek PhD and Prof.Huibert Mansvelder We used patch-clamp, optogenetics, and several other molecular approaches to investigate the corelease of Acetylcholine (ACh) and GABA in layer I and VI medial prefrontal cortex (mPFC). The aim of our research was to characterize and investigate the role of OCRL1, important protein in Lowe syndrome, in the brain. To do this, we used gene-gun transfection, immunostaining, 2-photon microscopy, and AMPAR trafficking assay in hippocampal neurons. These experiments were done in Department of Cognitive Neuroscience under supervision of Wei Ba and Nael Nadif Kasri The aim of our research was to characterize and investigate the role of OCRL1, important protein in Lowe syndrome, in the brain. The aim of this study was to determine candidate genes from chromosome 21 (HSA21) for Hirschsprung disease (HSCR) development in Down syndrome patients. As such, we systematically used microinjection of mRNA overexpression in zebrafish. This project was under supervision of Rajendra K. Chauhan and Prof. Robert Hofstra.
