DPhil (PhD) in the Systems Approaches to Biomedical Science Industrial Doctorate Centre, based in the Computational Biology group in the Department of Computer Science, University of Oxford.DPhil thesis: "Uncertainty characterisation in action potential modelling for cardiac drug safety".We use Bayesian statistical models to characterise uncertainty in mathematical models of heart cell bioelectrical behaviour and in drug screening data. These uncertainties are propagated into heart cell model predictions of how a particular drug will affect the cell's bioelectrical behaviour. We obtain a range of possible responses with associated probabilities, allowing for better-informed decision making when assessing a drug's cardiac risk.Data visualisation and statistical algorithms are implemented in Python which interfaces with the Chaste C++ library.

I spent two months in Liudmila Polonchuk's lab performing manual patch clamp experiments on stem cell-derived cardiomyocytes (SCCMs).I was recording action potentials (APs), both stimulated and spontaneous, in membrane voltage from SCCMs. APs were recorded in control solution and then in increasing concentrations of ion channel-blocking compounds.We fitted a range of mechanistic AP models to our recorded AP data within a larger statistical model to quantify some of the uncertainty associated with such computational modelling. Together with uncertainty quantification in ion channel screening data, we attempted to propagate uncertainty into larger-scale predictions of heart-compound interactions.
