      Continuing with the skills acquired in my summer internship at MCRI, I worked on a research project that was investigating the role of a new promising gene that could cause a neurodevelopmental disorder. The project had two aspects, functional assays, and in-silico analysis to determine the gene pathogenicity. In functional assays, I identified and analyzed single-gene mutations in patient cells, followed by the protein analysis and gene expression analysis. I used Confocal and Fluorescence Microscopy to determine the subcellular localization of the protein. Lastly, I conducted in-silico analysis using various bioinformatic tools that conclude and determine the possible gene pathogenicity.       The Brain and Mitochondrial division, Genetics-theme lab at MCRI investigates the genetic causes of neurological diseases along with mitochondrial diseases. As a research intern, I was trained in handling and maintaining a tissue culture lab. This included cell culturing and preservation of cell lines (HEK293 and Fibroblasts).       Working in the genetics-based laboratory in ICAR-CIFE, I worked on a project that focused on generating DNA barcodes of selected shellfish and fish from the Indian coast. Our group standardized the DNA isolation procedures for Barnacles and native fish species and optimized the PCR conditions for the COI gene from the isolated DNA. The COI gene sequence was then analyzed using Bioinformatics tools for developing the barcodes, which were successfully uploaded to Barcode of Life Data System, Centre for Biodiversity Genomics, Canada.
