• Independently conducted the isolation of memory B cells from specimens collected from HIV-positive human and non-human primate vaccine recipients by fluorescent-activated single-cell sorting. • Independently performed high-throughput cultures of primary memory B cells and high-throughput functional screenings. • Compiled, analyzed and summarized experimental results in both tabular and graphic forms, and Draw experiment conclusions provided to PI for further discussion. Trained new team members in general laboratory techniques and operation of equipment. • Assisted with, trained and supervised new employees for the laboratory high throughput sample pooling of the Duke MASC Program. • Through troubleshooting, improved performance, and reliability of Memory B Cell culture screening ELISA assay was obtained. • Optimized conditions and performed ELISAs to characterize Y. pestis-specific antibodies from immunized mice. • Achieved high throughput and yield of produced monoclonal antibodies by executing transient transfections, purification and quality control SDS/PAGE and Western blots. • Optimized conditions and performed immunoprecipitations to demonstrate the ability of glycan-reactive HIV-1-specific broadly neutralizing antibodies to cross-react with HLA-DR expressed on human B cells. • Supported laboratory organization by writing and updating SOPs, and supported data provenance goals by keeping highly detailed records of all experiments • Produced accurate budget information in a timely fashion to support grant and contract proposals. • Assisted the PI in preparing figures and methods sections of manuscripts, grants and presentations. • Prepared and presented a poster at a scientific meeting. • Efficiently maintained laboratory supply inventory and managed ordering to ensure sufficient stocks. • Knowledge of operations to numerous automation technology including Biomek liquid handling automation systems, and Biomek plate washers,.
