I worked on two proteins that play vital roles in the eukaryotic FeS cluster biogenesis pathway. I cloned constructs, expressed and purified proteins and characterized them using biochemical and biophysical techniques. I cloned constructs, expressed and purified proteins and characterized them using biochemical and biophysical techniques. My goal was to characterize these proteins and study the sulfide delivery mechanism in the biogenesis pathway. My accomplishments were: - I designed and developed protocols to express and purify two proteins using affinity, SEC and ion exchange chromatography. I also tested and validated biochemical and biophysical assays for the characterization of Drosophila cysteine desulfurase and its partner. - I established titration conditions for protein-protein interaction studies using Isothermal titration calorimetry - I used Circular dichroism to study protein folding changes in different constructs Areas of expertise I have gained expertise in traditional cloning, PCR, primer design, restriction digestion analysis, transformation, sonication, French pressure cell lysis, protein expression, protein detection using western blotting, protein purification using GE Healthcare AKTA-FPLC (gel filtration, ion exchange and affinity chromatography) and traditional bench columns, protein concentration estimation and protein-protein interaction studies using Circular dichroism and Isothermal titration calorimetry.
