At CIC Bio Gune I executed a project to develop a novel aptamer-guided method for targeted drug delivery in hepatocytes, troubleshooting all technical aspects of the Cell-SELEX methodology, which was designed and employed to discover aptamers capable of specifically recognizing the hepatic bile acid transporter/receptor NTCP. In addition, together with other colleagues, I worked on studying the transcriptional regulation of a transporter protein related with liver cancer and NAFLD/NASH, using DNA pulldown, proteomics and functional assays. Keywords: Project planning and experimental design, BSL2, mammalian cell culture lines, FACS, fluorescence microscopy, cell assay development, gene silencing, gene expression, in vitro assays, proteomics, NGS, aptamers, Cell-SELEX, adenovirus, lentivirus packaging Show less

At IIS La Fe I aimed at understanding the molecular mechanisms by which augmentin (amoxicillin-clavulanic acid) and anabolic steroids cause disruption of molecular metabolism of bile acids in the liver and drug-induced liver injury in susceptible individuals, as well as the mechanistic links between gut microbiota and NAFLD (https://goo.gl/rhKHSF). Keywords: primary hepatocytes, drug-induced liver injury, cholestasis, drug transporters, nuclear receptors, adenoviral based protein overexpression, bile acids, anabolic androgenic steroids, toxicogenomics, toxicology.
