  To understand the mechanism of action of GPCR’s using A2A and A2B adenosine receptor (AdoR) over expressing HEK-293 cell line. • The cells were cultured and maintained for functional assays. • Plasma membranes were extracted by sonication and ultracentrifuge technique. Radioligand binding assays were conducted to evaluate the binding potencies of different in-house compounds. • Cell based functional assays conducted using HTRF based cAMP assay to determine the dose response of selected agonists, to evaluate the compounds for their antagonistic activity on the receptors. • Schild plot analysis was carried out to characterize the mechanism of action of selected compounds.
