Uncovered how the Na/K pump interacts with other ion transport mechanisms to provide an adaptive advantage to extremely low salinities and the functional importance of the functional modifications induced special Na/K pump versions to the animals (zebrafish) that carry them. Determined that isoform ATP1a1a.5 (upregulated in extremely low salinity) has reduced protein function when compared to ATP1a1a.1, due to the 6 substitutions interacting with ions in the ion binding sites. I utilized an array of electrophysiological (TEVC), molecular modeling (Pymol/Alphafold), and biochemical techniques to discover these findings and validate my hypothesis.Mastered molecular biology techniques (gel electrophoresis, gene cloning, site directed mutagenesis, DNA/RNA purification, protein expression in oocytes and HEK cells) and trained multiple lab members to become proficient in each technique.
