Confirmed a highly protective effect of the large surface protein in prophylactic immunization against hepatitis B virus (HBV)-related woodchuck hepatitis virus (WHV) in a woodchuck model.• Established new and efficient in vitro neutralization assay using the primary woodchuck hepatocytes (PWH). Proved the high neutralization potential of antibodies produced against WHV large surface protein in the established assay.• Identified the most important sequence within WHV large surface protein which elicits antibody response associated with protection against WHV infection. • Created a new strategy for the improvement of HBV prophylactic and therapeutic vaccines’ immunogenicity: the combination of highly immunogenic non-infectious HBV core protein (HBcAg) capsid-like particles as carriers and immunogenicity enhancers for the neutralizing epitopes of the large HBV surface protein participating in binding to hepatocytes.• Proved the potency of HBcAg to induce a vigorous T- and B-cell immune responses and its ability to ensure a high level of B-cell immunogenicity to the fused surface protein’s sequence.
