Niyanta is all you could ask for as a co-worker and teammate! I have worked alongside Niyanta for over four years in the Pharmaceutical Sciences graduate program at UW-Madison. We have collaborated on a number of projects, one of which utilized scanning electron microscopy of brain tissue. I am continually impressed by her drive to move a project forward, seek the best solutions, and implement them fast. In particular, Niyanta’s communication and presentation skills are exceptional, allowing her to effectively communicate with investigators, other colleagues in the field, co-workers, and junior students she has mentored.       Drug delivery to the central nervous system (CNS) is challenging due to the presence of the blood-brain barrier (BBB) and blood-cerebrospinal fluid barriers (BCSFBs). My research in the laboratory of Assistant Prof. Robert Thorne focuses on investigating the DMPK of antibody based therapeutics following intranasal or vascular (systemic delivery). The intranasal pathway provides a non‐invasive route for drug delivery to the CNS which has implications for improving patient compliance and reducing costs. Using radioactivity and fluorescence based methods I have demonstrated that nasal delivery allows drugs access to perineural and perivascular pathways that potentially bypass the BBB. This research has: • Delineated the central nervous system distribution, dose response, and pharmacokinetics of radiolabeled IgG after intranasal and vascular dosing to assess the feasibility and saturability of each drug delivery route. • Investigated the effect of protein size on central nervous system distribution by comparing intranasal delivery of antibody fragments (Fab and single domain antibodies) versus full length IgG. • Developed a patented strategy to improve CNS delivery of intranasally applied antibodies by modulating nasal epithelial permeability using a physiologic enhancer - Matrix Metalloproteinase-9 (MMP-9) • Delineated that perivascular distribution is KEY to the nasal drug delivery route scaling across species • Provided the FIRST report for a 3 step nose‐to‐brain transport pathway for antibodies following intranasal delivery.
