My project focused on understanding the mechanisms by which synthetic STAT6-IP (designed as a potential therapeutic for allergic asthma) provides protective immunity in murine models of maladaptive Type 2 inflammation and Th2 adaptive immunity in the lung. I performed the various treatments for each mice group, as well as the subsequent lung collection and digestion. I then performed qPCR, ELISA and flow cytometry analysis to identify which neuropeptide(s) are induced by STAT6-IP in response to IL-33 delivery to the lungs and to clarify how STAT6-IP delivery after IL-33 interacts with this peptide superfamily to reduce ILC2 responses. I performed rRNA FISH on early embryos to delve into complex relationship between transcription and membraneless organelles. I also conducted timecourse experiment using a perfusion system and fluorescence microscopy to investigate the spatial distribution of RNA polymerase in E. coli. During Summer 2020, I developed image analysis pipeline to analyse E. coli transcription foci which demonstrate problem solving and critical thinking skills.
