My project is to explore type one Von Willebrand disease using mice models. In type one Von Willebrand disease the amount of Von Willebrand factor in the blood is decreased. A similar pathology is observed in mice due to abnormal expression of the B4galnt2 gene.Instead of being expressed in vascular endothelium (as in normal mice) Von Willebrand factor is expressed in intestinal epithelium. This leads to abnormal post-translational modifications that result in the early clearance of the protein from the peripheral blood. I will be focusing on the intergenic region that controls the expression of B4galnt2 in order to pinpoint the exact location of mutations responsible for the atypical expression. Show less
