Transformed the newly created mutant plasmid in Escherichia coli cells and expressed protein. Purified the protein tagged with six-Histidine residues in the C-terminal region using Talon (Co++) resin by affinity based column chromatography. • Subjected the proteins to mechanistic studies involving enzymatic assays to test protein encoded endonuclease activity, reverse transcriptase activity and target DNA/RNA binding ability. Visualized results using polyacrylamide Electrophoretic mobility shift assay (EMSA) and urea Denaturing gel. • Constructed and analyzed a homology based 3D model of the protein using bioinformatics tool. • Designed primers and oligo, used PCR and annealing method to construct double stranded and holiday junction DNA. Radiolabeled the DNA constructs with Phosphorus-32 (32P) isotope. • Optimized IPTG induced recombinant protein expression protocol and affinity chromatography based protein purification protocol to enhance the protein yield. • Gained expertise in managing multiple research projects involving research plan development, literature review, design & execution of experiments, optimization of protocols, data analysis; and participate in writing grant proposals and manuscripts. • Experience in laboratory inventory management and budgeting by taking responsibility for lab reagents ordering; material storage; and proper disposal of dangerous chemicals including radioactive materials • Served as a mentor to four graduate students and two undergraduate students providing training for research work • Maintenance and troubleshooting of lab equipment and training new users from different departments • Experience of wet laboratory systems including radiation handling station, and fume hood • 1 article in peer-reviewed journal and co-author of five oral and poster presentations
