Researcher at CNR in the field of neuroscience and brain physiology. Extensive research was performed in order to investigate how do inhibitory GABAergic interneurons (INs) (eg. Parvalbumin and Somatostatin INs) regulate cerebral blood flow in their interaction with glial cells astrocytes. In the living mice and brain slice preparations, optogenetics (Channelrhodopsin), electrophysiology and single/2-photon laser scanning microscope Ca2+ imaging techniques were performed to study Ca2+ signal dynamics in neurons and astrocytic processes and endfeet expressing genetically encoded Ca2+ indicators, expressed using AAV strategy injections. PCR genotyping, Immunocytochemistry, mice perfusions were also performed. Data analysis and coding using MatLab, During this period I participated in numerous international Conferences disseminating results of my research as an invited speaker. In the field of breast cancer research, I was testing whether the olive extracts could reduce and inhibit growth of breast cancer cells in vitro. During this 6 month research internship I utilised different experimental techniques, especially those used in cancer research. Cell culture of normal epithelial cells and breast cancer as well as ovarian cells was used. Work with a cellular infection model that was recently established in laboratory of AMP group. The model was used to test various compounds with the capacity to induce antimicrobial peptides (AMPs) and other host defence effector systems (reactive oxygen species, nitric oxide and autophagy). Infection of the model with bacteria and fungi and evaluation of the effect of different AMP-inducers alone and together with conventional antibiotics and anti-fungal treatment. Recently,promising results were obtained for one such inducer against multi drug resistant Klebsiella, which lend support to the feasibility of the project.
