  Supervisor: Prof. Giuseppe SacconeProject description (15 months): Studying and understanding the molecular pathway of sex determination in embryonic development stages. I focused my attention on RNA extraction from single Ceratitis capitata embryos as well as on quantitative expression analysis by using the SYBR Green real-time PCR technique. Moreover, DNA extraction, RNA extraction, complementary DNA (cDNA) synthesis, sequencing, cloning and plasmid purification are a part of my knowledge. On the other hand, I also learnt in vivo experimentation using the CRISPR/Cas9 system in the MedFly thanks to the micro-injection technique. During this period, I performed in vitro single guide RNA (sgRNA) synthesis in order to use CRISPR/Cas9 as a pre-assembled ribonucleoprotein complex. In such a context, downstream analyses of the ON-target of CRISPR/Cas9 system, such as insertion and deletion (InDel) detection, were carried out. Lastly, I was introduced to the basis of insect facility rearing.

First marking experiment and release of sterile males of Aedes albopictus on the island of Procida for the calculation of the density per hectare of this species.Acquired competencies: principal monitoring methods of the Asian tiger mosquito Aedes albopictus using ovitraps, BG-Sentinel-2 traps, and Human Landing Catches (HLC).
