      Performing under Dr. Hans Schelvis Abstract - Our research involves spectroscopic analysis of an irradiated thymine solution to characterize the stereoisomeric dimers that form after being exposed to UV light. The dimerization that occurs when thymine bases are exposed to UV light can cause structural changes in DNA and can inhibit replication. To form these dimers in the lab, we exposed a thymine solution to UV light for two hours. This solution was than dried and analyzed by a few techniques to determine the damage caused by irradiation. High-performance liquid chromatography (HPLC) was used to separate and purify each peak on the chromatogram which is speculated to be different stereoisomers. These separate peaks will be analyzed by nuclear magnetic resonance (NMR) coupled with Fourier transform infrared (FTIR) and Raman spectroscopy to determine the structural differences between them. The structural differences of these stereoisomers will also affect their ability to be repaired by the DNA repair enzyme photolyase, the dimers will be tested for this as well. Show less
