Designed and automated in vitro assays to support hit-to-lead identification of small molecule compound protein-protein interaction (PPI) inhibitors - Developed biochemical assays, such as AlphaLISA and MSD ELISA, starting from identifying reagents to completing the protocol of high quality, robust assays. Facilitated the transfer of such assays to CRO and troubleshooting efforts. - Developed cellular assays to assess compound inhibition of cell binding to protein covered wells, and ran weekly screens of developed small molecule compounds through the assays - Validated CRISPR KO cell line gene expression via qPCR with a Quantstudio 6, and via FACs with a Sartorius iQue3 - Automated biochemical and cellular assays for high throughput screening utilizing liquid handlers and plate washers integrated on a robotics system from HighRes Biosolutions, and included instruments such as Multidrop Combii, Echo Acoustic Dispenser, Agilent Bravo, Biotek EL406 Plate Washer, BlueCatBio BlueWasher - Acquired assay data with a PherastarFSX, and performed analysis with GraphPadPrism and GeneData Screener 18 Designed in vitro assays to assess downstream signaling of GPCR targets to support drug discovery efforts and SAR studies of developed peptide and small molecule compounds - Transfected GPCRs and developed stable cell line clones in adherent cell cultures utilizing FugeneHD kits - Developed cAMP HTRF assays using Cisbio kits to assess stimulation of transfected receptors with test compounds - Developed imaging-based receptor internalization assays using an ImageXpress Micro Confocal Imager Show less
