Optimization of tissue-staining methods (IHC and IF) on specimens used for DNA retrieval by three main steps: Laser Capture Microdissection (LCM), followed by Proteinase K (PK) digestion and DNA purification. Three-dimensional mapping of tumor progression in prostate cancer by use of LCM on varying layers of human prostate tissue, followed by CGH array for DNA methylation patterns in the stroma. Optimization of IHC using CD45.2 as a cell marker for early-stage acute lymphocytic leukemia (ALL) in mouse bone marrow (BM). Using IHC at different time points of tumor development to visualize ALL-promoting niches and ALL progression in the BM. Detection of thymic stromal lymphopoietin (TSLP) in mouse BM. Measuring relative gene expression levels of Tslp by Real-Time PCR on cDNA samples transcribed from purified RNA in mouse BM samples. Show less
