Design of a versatile metabolic chassis for Oligosaccharides production in E. coli.Teaching: bachelor's degree, biology, biochemistry and biotechnologies - UFR Pharmacie and Université Joseph Fourier.OLIGOMET aims at developing a versatile metabolic chassis for efficient production of added-value oligosaccharides. The chassis will be designed by using E. coli as platform organism and system-level approaches for metabolic optimization. The versatility of the metabolic chassis will be evaluated by plugging two types of native or engineered carbohydrate active enzymes, to produce oligosaccharides of high added-value.

Study of the relation between the putative flippase ALA10 and the galactolipids synthesis in Arabidopsis thaliana. Work was mainly done on analysis of interesting gene transcripts from galactolipids synthesis by quantitative PCR.I worked on various A. thaliana lines, WT and mutants, and used culture media variation to identify the possible relation between AlA10 and galactolipids synthesis. I completed transcripts analysis by protein analysis. It allowed us to obtain a first proof of the existence of a relation between putative ALA10 flippase and galactolipids synthesis.

Study of protein responsible of small RNA (21nt) generation by cleavage of double stranded RNA in Arabidopsis thaliana. Thanks to technics allowing protein-protein interaction study, coupled withdetection technics, it has been possible to study DRB4, a double-stranded RNA binding protein involved in double-stranded RNA cleavage by a protein dicer-like DCL4.I mainly used pull down assay, immunoprecipitation and western blot. Analyse de l'influence de phospholipides sur la fixation de la protéine MGD1 à des liposomes dans le cadre de la régulation de la synthèse des galactolipides chez Arabidopsis thaliana. Dans ce travail, des techniques de surproduction et de purification de la protéine MGD1 ont été appliquées. Puis la création de liposomes de composition lipidique déterminée nous a permis de tester l'attachement de MGD1 à trois phospholipides différents. Les limites de cette méthode ont été discutées et des alternatives ont été proposées.
