      In the Department of Molecular Microbiology I have learned several key techniques for working with Streptomyces. These techniques include the cultivation of Streptomyces and the preparation of spore samples for long-term storage and microscopic analysis (image acquisition and analysis using the ImageJ software). I prepared spore stocks of several different Streptomyces mutant strains (expected to be affected in the formation of regularly-sized spores) and used phase contrast microscopy to acquire images of these different spore stocks. I subsequently analysed these images using the ImageJ software and determined that some of Streptomyces mutant strains were clearly affected in making spores of equal size. I also obtained training in automated measurement of Streptomyces spores using the Coulter Counter Multisizer 4e. This devise allows the automated and highly quantitative measurement of particle sizes, such as spores produced by Streptomyces. I used the Coulter Counter to determine the spore size distribution of spore stocks and used this data to complement the results obtained from the microscopic image analyses of spores. Show less
