Working in the ReproTracker group it is our goal to test customer compounds on teratogenicity, using hiPS cells and differentiating them into three lineages, which qPCR as a read out. I am on the R&D part of this which involves optimising the existing protocols, improving throughput (automation), developing new differentiations to extend the service and collaborate with other companies who would like to test their compounds without using in vivo models. Besides the execution of protocols, I also design, plan the experiments and analyse the data in cooperation with the senior scientist. Other duties are training new team members and general lab work such as keeping track of the stocks, autoclaving tips and cleaning the lab on a regular basis. Show less

I was the only technician for 3 different groups, helping PhD students with their project and had my own small projects. Some of my tasks were cloning, culturing of mammalian cell line such as mES and iPS, smFISH, setting up ClampFISH, IF stainings/labeling and cryosectioning. Besides the lab work I was also responsible for maintaining the storage room, ordering products and keeping the lab clean, tidy and safe. My main task here was to receive the blood and urine samples and prep them for shipments to other labs for analysis. This also meant to prep for different studies in terms of how many tubes of what kind is needed, where do the samples need to be shipped to and plan the shipments. For the blood samples we mainly had to take either the plasma or serum. For one study we also had to isolate cells using RoboSep, count the cells using a MACS and isolate DNA. During this internschip I was searching for and sequencing genes that might be involved in migration disorders in the brain. This also includes designing and testing primers.
