The project aim was to construct an in vitro model of a 3D neuronal network, combining layered biomaterials, neural cells, stimulation and monitoring. My main focus in this project was to study the development of neural networks by using human pluripotent stem cells (hESCs and hiPSCs). More specifically, I studied the role of astrocytes in developing neural networks, and how different environments contribute to the network development. I also optimized adherent differentiation protocol for neural cells, tested different biomaterials for neural cells, and was involved in developing a novel 3D MEA. This project led me to travel to project meetings in Europe and to visit collaborators’ laboratories. I also contributed to the coordination of our group’s research, managed collaboration with the project partners, and prepared milestone reports. In addition I familiarized new employees and mentored students in their own projects. The main methods I used were microelectrode array (MEA) for studying neural network activity, immunocytochemistry for protein expression, microscoping for monitoring cell growth and morphology, and FACS (fluorescence activated cell sorting) and MACS (magnetic-activated cell sorting) for sorting neuronal cells. The methods I used were microelectrode array (MEA) for monitoring cell activity, immunocytochemistry for protein expression, and Live/Dead for studying cell viability. This collaboration project taught me the key aspects of fruitful collaboration, how to work in interdisciplinary setting, and most of all the importance of unambiguous communication.       I worked as a summer trainee in Johanna Schleutker's cancer genetics group. I performed different laboratory analysis from patient samples. Extracted DNA from whole blood, did PCR analysis and genotyping (with sequencing and TaqMan-chemistry), screened mutations with HRM (High Resolution Melting) technique, and cultured lymphoblasts.
