      Study of the plant-pollinator relationship in order to clarify the role of floral rewards - in particular the quality of the nectar - on attraction and behavior of insects, and consequently on the plant's reproductive success and wild population's fitness. During the internship, the field study involved observation, sampling and identification of insects pollinating the species Gentiana lutea L., while in the laboratory quali-quantitative surveys were conducted on floral rewards (nectar, pollen), with particular attention to the amino acid component. They were carried out tests of preference and behavior tests on Bombus terrestris and Apis mellifera colonies. In collaboration with the Department of Life Sciences of the University of Siena, samples of nectar and hemolymph were analyzed by HPLC chromatography to identify protein and non-protein amino acids.       The project tested the reliability of RAPD-PCR technique to evaluate its possible use in environmental monitoring projects. In particular, the internship focused on the different species of the genus Barbus in Italy. Thanks to the RAPD method should be possible to identify immediately members of a given species (due to the presence of so-called diagnostic bands), to evaluate the extent of their presence in the habitat, to discriminate between pure and hybrid individuals and to proceed accordingly. The activities carried out during the internship were: DNA extraction using phenol-chloroform method, gel electrophoresis and size calculation of the individual bands. In addition, the construction of a phylogenetic tree and the assessment of genetic distances through TFPGA program were carried out. Show less
